Description
Thermo-Start Taq DNA Polymerase is engineered to provide maximum sensitivity and specificity for PCR. The chemically-modified Thermo-Start Taq remains completely inactive until it is ‘switched-on’ by a high temperature incubation step at the start of the PCR reaction. This prevents non-specific amplification during reaction set-up, eliminating smearing and the formation of non-target bands.
To protect the enzyme during this 95°C/15 minute activation step, a proprietary buffer is supplied with the enzyme. This ensures an optimal response from the enzyme once it has been activated, resulting in higher yields than those obtained with systems based on chemical modifications alone. When other enzymes have completely lost all activity due to high denaturation temperatures Thermo-Start is still at least 50% active. This thermal stability allows for the high temperature denaturation steps used when working with complex targets.
The combination of Thermo-Start Taq and optimized buffers increases sensitivity and specificity without any compromise on yield. The hot-start system also increases PCR efficiency by preventing amplification of primer dimers formed at low temperatures, preserving reaction components for template amplification.
A High Performance Buffer formulation is available to increase yield while still maintaining a highly sensitive and specific reaction. The High Performance Buffer works extremely well with complex and GC-rich templates.
