Verso 1-step kits provide a sensitive and convenient method for performing RT-PCR in a single reaction tube. The buffer system of the Verso 1-Step RT-PCR Kit has been specifically optimized to work across a wide range of RNA templates for both transcription and amplification. Special enhancers and additives have been used to generate a reaction mixture which allows both reverse transcriptase and DNA polymerase to work optimally.
The Verso RT Enzyme can be used at temperatures up to 57°C, avoiding problems caused by secondary structure in the RNA template. It also shows a better dynamic range than AMV or MMuLV. The Verso 1-Step RT-PCR Kit can detect as little as 10pg of total RNA or 1pg of mRNA. This allows for accurate detection and quantification of target RNA over a broad template range, ensuring that increases in target RNA concentration give rise to corresponding increases in PCR product.
The master mix format minimizes reagent-handling steps, thus reducing errors and the risk of contamination. It is supplied at a 2X concentration, providing extra flexibility for primer and template concentrations. The Verso RT Enzyme and RNase inhibitor are provided in a separate tube, allowing negative controls without RT enzyme to be carried out easily.

Verso 1-Step RT-PCR Kit with ReddyMix™
The Verso 1-Step kit is available in ReddyMix format, where the master mix contains an inert red tracking dye and gel loading precipitant. After thermal cycling, a sample of the reaction can be removed and loaded directly onto an agarose gel without the addition of loading dye. Combining the components for PCR and the gel-loading buffer in a single master mix eliminates many pre- and post- PCR set-up steps. ReddyMix Kits give the same high performance as standard Verso 1-Step Kits. The inert red dye also allows for easy sample visualization during reaction set-up.

Verso 1-Step Hot-Start RT-PCR Kit
The Verso Hot-Start Kit combines the proprietary Verso RT Enzyme and Thermo-Start Taq DNA Polymerase to provide maximum sensitivity and specificity. The chemically-modified Thermo-Start Taq remains completely inactive until a 95ºC incubation step at the beginning of the PCR reaction. This prevents non-specific amplification during the reaction set-up and the RT step, eliminating non-target bands.
The combination of the Verso RT enzyme, Thermo-Start Taq and optimized buffers increases sensitivity and specificity without any compromise on yield. The hot-start system also increases PCR efficiency by preventing amplification of primer dimers formed at low temperatures, preserving reaction components for template amplification.
The proprietary buffer formulation protects Thermo-Start Taq during high temperature steps, ensuring optimal performance throughout the PCR reaction. This thermal stability allows for the high temperature denaturation steps used when working with complex targets.
