Thermo Scientific SIEVE software is an automated software package for the label-free, semi-quantitative differential expression analysis of proteins, peptides and metabolites. It is a statistically rigorous tool for analyzing data from proteomic and metabolomic biomarker discovery experiments.
Product Detail
SIEVE™ software performs differential analysis of sample populations, comparing LC/MSn spectra from "healthy" and "disease" (or control versus treatment) samples to find differentially expressed features. Trend analysis can be performed to detect changes associated with dosage effects or time points, enabling comparison of multiple classes of samples. Peptides that show statistically significant differences can be searched against protein databases to determine peptide and protein identities. Or in the equivalent metabolomics workflow, metabolites of interest can be sent to ChemSpider for identification. Using SIEVE software to pre-filter data greatly reduces the number of spectra that need to be searched, significantly decreasing the time spent on identification and increasing the throughput of complex biomarker discovery experiments.
SIEVE software uses the MS intensities from raw LC/MS data to find statistical differences without the need to manipulate or "model" peaks
The SIEVE process is label-free – not requiring isotopic tags or labels of any kind
SIEVE software has the power to compare hundreds of LC/MS experiments at a time, but can also perform a simple comparison of two sample files.
SIEVE employs a novel algorithm called ChromAlign™ for chromatographic alignment prior to a proprietary iterative process called Recursive Base-Peak Framing to find differences that are statistically meaningful.
SIEVE determines a p-value for the expression ratio of each putative biomarker, providing an extra measure of confidence.
SIEVE accommodates high-resolution and low-resolution data generated on the Thermo Scientific LC/MS and GC/MS instruments, including data from the LTQ™, LTQ Orbitrap™, and Exactive™ product lines
Combining SIEVE software for label-free differential expression with the unique analytical power of high-resolution Thermo Scientific LC/MS systems creates complete solutions for biomarker discovery.
