The Neuronal Profiling image analysis module performs measurements that characterize neurite outgrowth such as neurite count, neurite total length, average distance of branch points and length of longest neurite (with and without branch points). It also allows you to define subpopulations of interest with the BioApplication Events Wizard, and the application characterizes these subpopulations accordingly.
Key Features:
-
Accurate tracing of very long neurites
-
Ability to select/reject neurites based on intensity, width or length
-
Validated for robust performance with primary neurons and neuronal cell lines
-
Events Wizard allows generation of morphological and /or expression profiles for easy screening
-
Ability to segment and analyze clumped neurons

Figure 1. Rat cortical neurons stained with the Neurite Outgrowth HitKit® HCS Reagent Kit and analyzed with the Neuronal Profiling BioApplication. Cell bodies are outlined in blue, branch/cross points in yellow, and neurites in aqua, green, and purple.

Figure 2. Dose-response of Neuroscreen-1 cells to increasing concentrations of NGF. Each column of a 96-well plate was treated with a different dose of NGF for 72 hours, fixed and stained with the Neurite Outgrowth HitKit® HCS Reagent Kit. Images were acquired and analyzed by the Neuronal Profiling BioApplication. Each data point represents the well-level average of 300 cells collected from 2-20 imaged fields. The well-to-well reproducibility is demonstrated by the overlapping dose-response curves for the feature Average Neurite Length.
