The sensitivity and selectivity of electrospray mass spectrometry, combined with the on-line sample concentration and desalting capabilities of reverse-phase HPLC, creates a tandem system of choice for the automated, high-throughput analysis of compounds in a mixture.
Conventional LC systems generally offer narrow/sharp chromatographic peaks for each eluting component. However, these sharp peaks also limit the MS analysis period of each peak to approximately 10 seconds, which sometimes is too short for a mass spectrometer to acquire comprehensive structural information or useful MS/MS data for co-eluting components of low abundance in a highly complex mixture. A novel technique, called “peak parking,” has been developed to overcome this limitation.
