Evaluation of ICAT Reagents for Protein Quantification

Methods: Mixtures of human serum albumin, human transferrin, and human amacroglobulin were processed using the cleavable ICAT method and the resulting labeled peptides analyzed by 1D LC/MS/MS.

Results: Quantification of proteins in simple mixtures using the ICAT labeling approach requires the presence of several highly reactive cysteine residues in the targeted proteins and allows consistent detection, identification and quantification by LC/MS/MS in simple mixtures of proteins present at similar concentrations.
