Tip of the Month: Optimizing H-ESI Source Conditions for FAIMS Analysis FAIMS analysis works best with dry ions. To ensure the ions are dry, the following H-ESI source probe positions need to be optimized:The user needs to ensure that the H-ESI probe is not spraying too close to the entrance plate opening. Typically, the probe is positioned more than 1.5 mm from the entrance plate opening. Usually, the farther back the probe the better, but placement is compound, mobile phase, and liquid flow rate dependent. Tuning is always started with the H-ESI probe in the C or D position, centered, and as far back as possible. The micrometer can be used to bring the probe closer to achieve the maximum stable signal.

Forensic Toxicology: Identification of Psychotropic Substances in Mushrooms and Chocolate by UHPLC/MS

The identification of controlled substances in food matrices is a challenge for forensic laboratories. Classical techniques, such as color tests and thin-layer chromatography, do not provide molecular structural information and cannot be used as a principal means of identification. Given the increasing emphasis on instrumental methods of analysis and the limitations of the traditional instruments in the field of forensics, another solution is necessary. This application note describes the method to develop a UHPLC/MS method for detection, separation and confirmation of psilocybin and psilocin in mushrooms and chocolate matrices.

Determination of Pesticide Residues in Processed Fruits and Vegetables According to EU Directive 91/414 EEC

By researchers at General Chemical State Laboratory, Pesticide Residues Laboratory, Athens, Greece

A diet rich in fruits and vegetables is thought to reduce the risk of some types of cancer, atherosclerosis, and heart disease. However, commercially grown produce often contains high levels of pesticide residues that can lead to serious health problems when consumed. Due in large part to growing public concern over the amount of pesticide residues in foods, the European Union (EU) has enacted several directives to fix Maximum Residue Limits (MRLs) for different pesticide residues in food of plant origin. In this application note a multi-residue LC-ESI-MS/MS method was developed for the reliable confirmation and quantification of pesticides from different chemical classes at low ppb levels in food matrices.

Bioanalysis: Paroxetine in Rat Plasma - Enhanced Reproducibility Performance of the TSQ Vantage

In May 2001, the FDA promulgated its first official guidance for bioanalytical methods. This document is titled Guidance for Industry Bioanalytical Method Validation, and serves as a guide for bioanalytical methods supporting clinical and non-clinical studies. Recently, the reproducibility and accuracy of the concentration determined in incurred samples was subject to significant debate, outlined in the American Association of Pharmaceutical Scientists (AAPS) journal paper titled Workshop/Conference Report - Quantitative Bioanalytical Methods Validation and Implementation: Best Practices for Chromatographic and Ligand Binding Assays.

Over the past three years the combination of Orbitrap technology combined with a linear ion trap has become an established platform for high resolution, accurate mass LC/MSn analysis. The high resolving power, mass accuracy and dynamic range of the Orbitrap analyzer allow rigorous characterization of complex mixtures even in the absence of precursor ion mass selection.
