Tip of the Month: Improving ETD Activation Times

For increased coverage of complex sample mixtures, the activation time used for electron transfer dissociation (ETD) fragmentation of higher charge state peptides should be around 50ms. However, upon tuning the ETD activation time for +3 and higher charge states, you may find that the optimized activation time is significantly greater than this recommended value. An easy way to reduce the activation time is to increase the reagent ion automatic gain control (AGC) target setting from its default value of 1.00e+05.

Bioanalysis: Using FAIMS to Remove Interference in a Validated Small Molecule Assay

Bioanalytical methods are developed to determine drug concentrations and must be validated using the same matrix as the study subjects. The ideal control matrix contains no interferences, but this must be tested during the validation according to FDA guidelines. High-Field Asymmetric waveform Ion Mobility Spectrometry (FAIMS) increases the selectivity of analysis by selecting which ions are allowed into the mass spectrometer. This technique filters out interferences and leaves the signal cleaner than LC-MS/MS alone.

Forensic Analysis: Analyzing Opiates in Whole Blood with Online Sample Extraction and LC-MS/MS

The opiate morphine, and its derivatives, are medicines often used for pain-relief, cough-relief and as anti-diarrhoeals. For example, codeine and dihydrocodeine (morphine derivatives) are available in over-the-counter preparations in combination with paracetamol (acetaminophen) and are slowly metabolized to morphine and dihydromorphine respectively. The forensic toxicologist is often asked to interpret results and possibly account for time of death in opiate (especially heroin) abuse cases. This task can be made easier if it is possible to identify and quantify the components such as 6-MAM, morphine, codeine, dihydrocodeine and the glucuronides in whole blood rather than urine.

Environmental: Case Study - Online Sample Extraction and LC-MS/MS for Water Analysis

The Environmental Analytical Chemistry Laboratory at the University of Montreal comprises approximately 20 researchers focused on finding the best analytical methods for environmental studies. The laboratory is currently collaborating with the City of Montreal, the Drinking Water Research Chair at the École Polytechnique de Montréal, and Environment Canada in order to identify and quantify pharmaceutical residues in the city's waste and drinking water. To conduct this research, the University has developed a new online sample preparation method using a customized Thermo Scientific EQuan system for solid phase extraction coupled to liquid chromatography and tandem mass spectrometry (SPE-LC-MS/MS) analysis.

Chromatography: Fast Analysis of Water-Soluble Vitamins

Water-soluble vitamins represent a group of various, essential dietary compounds, which are classified by their biochemical activities, not their physico-chemical properties. A single vitamin generally is formed by several vitamers (various chemical species, each of which displays the same biological activity). As a result of their different chemical properties, it can be difficult to reliably and simultaneously analyze vitamins. This application note describes a fast LC method for the analysis of water-soluble vitamins using a Thermo Scientific Hypersil GOLD aQ column.

Glycosylation of Asn, Ser or Thr is arguably the most common known post-translational modification (PTM), resulting in a multitude of highly heterogeneous protein isoforms. Traditional electrophoretic and CID LC-MS/MS approaches to analysis of glycopeptides have significant limitations. This application note demonstrates the advantages of a workflow that incorporates porous graphite chromatography and electron transfer dissociation (ETD) LC-MS/MS for identification and characterization of glycopeptides.

Trap Talk: Novel Dual-Pressure Linear Ion Trap Mass Spectrometer Offers Breakthrough Performance in Proteomics Experiments

This application note addresses the challenging analysis of a complex, multi-organ peptide digest of Caenorhabditis elegans (C. elegans) to assess the performance of the innovative new Thermo Scientific LTQ Velos mass spectrometer. Performance of this new instrument is benchmarked against the previous state-of-the-art linear ion trap and also against a well-known quadrupole time-of-flight mass spectrometer.
