 Dr. Alexander Makarov Receives 2008 ASMS Distinguished Contribution in Mass Spectrometry Award  
Dr. Alexander Makarov, Thermo Fisher Scientific's Director of Global Research for Life Sciences Mass Spectrometry, received the 2008 Distinguished Contribution in Mass Spectrometry Award from the American Society for Mass Spectrometry (ASMS). Dr. Makarov, who has authored more than 30 papers and holds four patents, received the award for his pioneering development of the Orbitrap™ mass spectrometer. The Orbitrap analyzer has succeeded in making high performance research analytical power routine. This has enabled ultra-high resolution mass spectrometry to be used to solve complex analytical problems such as those found in pesticide analysis and drug discovery.

 Fossilomics Research Enabled by Mass Spectrometry  
Fossilomics is the analysis of amino acid sequences, post-translational modifications and phylogeny of fossil bone proteins from extinct organisms. Fossilomics has given credibility to the theory that dinosaurs' closest living relatives are modern-day birds. Molecular analysis of a shred of 68-million-year-old Tyrannosaurus rex protein, along with that of 21 modern species, confirms that dinosaurs share common ancestry with chickens, ostriches, and to a lesser extent, alligators. Mass spectrometers, specifically the Thermo Scientific LTQ XL and LTQ Orbitrap were utilized in this breakthrough fossilomics studies.

 Tissue Imaging Mass Spectrometry  
Irinotecan, isolated from the Chinese tree Camptotheca acuminata, has shown strong antitumor activity against a wide variety of human solid tumor xenografts in mice. Irinotecan is a DNA topoisomerase I inhibitor that binds to and prevents dissociation of the DNA-topoisomerase I complex (involved in DNA replication), thereby inhibiting enzyme activity. Lack of microvessels has been shown to offer resistance to irinotecan therapy in human head and squamous carcinomas. This application note describes the method used to determine irinotecan metabolites and their distribution in human tumors directly from tissue by MALDI LTQ XL Imaging MS.

An integral part of drug discovery and development is the identification of drug metabolites formed through phase I and phase II metabolic reactions. These metabolites may have either intrinsic pharmacological activity or display specific toxicity.
